<p><span style="font-family:'Times New Roman', Times, serif;font-size:medium;">細胞由DNA/RNA、蛋白質、脂質和碳水化合物等生物分子組成,以執行其功能,但我們對細胞結構與功能的理解主要集中在DNA/RNA與蛋白質上。然而,隨著基因剔除小鼠(knock-out mice)、小干擾RNA(siRNA)及質譜分析(MASS spectrometry)等技術的進步,情況逐漸改變,使得脂質組學(lipidomics)與醣組學(glycomics)的結構與功能研究成為可能。</span><br /></p><p data-start="197" data-end="367"><span style="font-family:'Times New Roman', Times, serif;font-size:medium;">在本課程中,我們將介紹相關的歷史背景,並探討數種在生物上具有重要意義的醣類共軛物(glycoconjugates),如N-醣鏈(N-Glycans)、O-醣鏈(O-Glycans)、糖鞘脂(Glycosphingolipids)、蛋白聚醣(Proteoglycans),以及最新發現的醣-RNA(glycol-RNA)的結構基礎與功能角色。</span></p><p data-start="369" data-end="409" data-is-last-node="" data-is-only-node=""><span style="font-family:'Times New Roman', Times, serif;font-size:medium;">課程後段將透過近幾年的研究文獻,探討醣類在細胞訊號傳遞、發育及人類疾病中的作用。</span></p><p><br /><span style="font-family:'Times New Roman', Times, serif;font-size:medium;color:#000000;">Cell consists of biomolecules of DNA/RNA,protein, lipid and carbohydrates for its function, but our understandings of cellular structure and function have been mainly focused on DNA/RNA and proteins. Recent technological advances in knock-out mice, siRNA and MASS spectroscopy, however, have gradually changed the situation to allow the structural and functional characterization of lipidomics and glycomics. In this course, we will provide the historical background and evaluate the structural basis and functional role of several biologically important glycoconjugates such as N-Glycans, O-Glycans, Glycosphingolipids, Proteoglycans and the most recent glycol-RNA. Issues relating to the role of glycans in cell signal/development and human disease will then be discussed by using literatures available within the last couple years.</span> </p>
